چکیده

Phosphatidylglycerol has been shown to be a constituent of the lipids of both plant and animal tissues by Benson and Maruo (1) and Strickland and Benson (2). It has also been detected in bacteria (3,4). Benson and Maruo (1) have called attention to the rapid rate of metabolism of phosphatidylglycerol in plants, and Kanfer and Kennedy 1 have obtained evidence that this phosphatide is a metabolically active component of the lipids of Escher&u coli. Recently, Macfarlane has reported (5) that phosphatidylglycerol may be the phospholipid moiety of certain lipid-amino acid complexes in Clostridium welchii. The metabolism of phosphatidylglycerol thus appears to assume growing importance, but little has been known about detailed enzymatic pathways for its biosynthesis. In the present paper, evidence will be presented for the occurrence of the following reactions, catalyzed by enzymes from liver, leading to the formation of phosphatidylglycerol.

N-Acetylsphingosine stimulates phosphatidylglycerolphosphate synthase activity in H9c2 cardiac cells.

Cardiolipin and phosphatidylglycerol biosynthesis were examined in H9c2 cells incubated with short-chain ceramides. Incubation of cells with N-acetylsphingosine or N-hexanoylsphingosine stimulated [1, 3-3H]glycerol incorporation into phosphatidylglycerol and cardiolipin, with N-acetylsphingosine having the greater effect. The mechanism for the ceramide-mediated stimulation of de novo phosphatid...

The pgp1 mutant locus of Arabidopsis encodes a phosphatidylglycerolphosphate synthase with impaired activity.

Phosphatidylglycerol is a ubiquitous phospholipid that is also present in the photosynthetic membranes of plants. Multiple independent lines of evidence suggest that this lipid plays a critical role for the proper function of photosynthetic membranes and cold acclimation. In eukaryotes, different subcellular compartments are competent for the biosynthesis of phosphatidylglycerol. Details on the...

Phosphatidylglycerol biosynthesis in chloroplasts of Arabidopsis mutants deficient in acyl-ACP glycerol-3- phosphate acyltransferase.

The biosynthesis of phosphatidylglycerol represents a central pathway in lipid metabolism in all organisms. The enzyme catalyzing the first reaction of the pathway in the plastid, glycerol-3-phosphate acyl-acyl carrier protein acyltransferase, is thought to be encoded in Arabidopsis by the ATS1 locus. A number of genetic mutants deficient in this activity have been described. However, the corre...

Participation of aminoacyl transfer ribonucleic acid in aminoacyl phosphatidylglycerol synthesis. II. Specificity of alanyl phosphatidylglycerol synthetase.

The biosynthesis of lysyl phosphatidylglycerol by a particulate enzyme from Staphylococcus aurelis involves the transfer of a lysyl group from lysyl-transfer RNA to phosphatidylglycerol. S-p-Aminoethylcysteinyl-tRNA’Y”, prepared enzymatically from the lysine analogue, S-p-aminoethylcysteine, serves as an effective substrate in the enzymatic formation of aminoethylcysteinyl phosphatidylglycerol....

Cardiolipin biosynthesis in the isolated heart.

The pathway for the biosynthesis of new cardiolipin was investigated in the isolated perfused intact rat heart. Isolated rat hearts were perfused in the Langendorff mode for up to 60 min with Krebs-Henseleit buffer containing 0.1 microM [U-14C]glycerol. Analysis of radioactivity incorporated into phospholipids in the organic phase revealed an increase in radioactivity incorporated into phosphat...
